Methods: In the rectal biopsy specimens from a patient with ulcerative colitis, detection of Epstein-Barr virus and human herpesvirus 6 was performed by the single strand PCR method. To localize Epstein-Barr virus in the paraffin-embedded biopsy specimens from hemorrhagic inflamed rectal mucosa and faintly inflamed terminal ileal mucosa, we attempted to detect Epstein-Barr virus-encoded small RNA-1 by in situ hybridization and Epstein-Barr virus latent membrane protein-1 by immunohistochemical staining. Results: Epstein-Barr virus and human herpesvirus 6 were detected in the rectal biopsy specimens by the single strand PCR. Positive signals for Epstein-Barr virus-encoded small RNA-1 were recognized in nuclei of some epithelial cells from both the rectum and the terminal ileum. Positive staining for Epstein-Barr virus latent membrane protein-1 was detected in cytoplasm of some epithelial cells and mucosal lymphocytes of the rectal and the terminal ileal mucosa. Conclusions: This report describes a patient with ulcerative colitis, the onset of which coincided with active Epstein-Barr virus and human herpesvirus 6 infection. This is the first demonstration of Epstein-Barr virus in the epithelial cells of the human colon by in situ hybridization and immunohistochemical staining. ----
